Descripción del caso clínico. El 90 % de una parvada de 800 pollos criollos de tres semanas de edad murió en 72 horas. A partir de cinco pollos enfermos que presentaron signos de depresión severa, caquécticas y con secreción serosa mucosa, nasal y ocular, se realizó el diagnóstico integral que sugería una enfermedad aguda de alta mortalidad como la enfermedad de Newcastle o influenza aviar de alta patogenicidad.

Hallazgos clínicos. Se observaron hemorragias en la tráquea, el proventrículo y las tonsilas cecales, pero en el estudio histopatológico, se reveló una necrosis y hemorragias en la médula ósea, los timos, el bazo, la bolsa de Fabricio, las tonsilas cecales, la tráquea y los bronquios. En la médula ósea, se advirtió también una hipoplasia hematopoyética severa con depósito de material acidofílico multifocal y necrosis celular multifocal. En el timo, el bazo y las tonsilas, se encontró una depleción severa de linfocitos, mientras que en la bolsa de Fabricio, los folículos linfoides estaban muy atrofiados y la Bolsa tenía aspecto adenoide con proliferación del tejido conectivo fibroso e infiltrado difuso de linfocitos. En algunas folias, donde aún se encontraban folículos linfoides, se observó una gran necrosis multifocal de los linfocitos. El conjunto de lesiones macroscópicas e histológicas sugerían una enfermedad hemorrágica y necrótica sistémica de curso agudo que sustentaban el diagnóstico de influenza aviar y enfermedad de Newcastle.

Pruebas complementarias de laboratorio. El pulmón y la tráquea fueron los tejidos con las lesiones macroscópicas más significativas. Únicamente el virus que se aisló de la tráquea y se analizó por RT-PCR dio positivo a la enfermedad de Newcastle. La secuenciación de nucleótidos del gen F mostró una similitud del 97 % con la secuencia de la cepa México/01/10 (JX974435) del virus de la enfermedad de Newcastle. Además, ambos virus comparten los mismos cuatro aminoácidos básicos en el sitio de corte de la proteína F (RRQKR*F) que los clasifica dentro de los patotipos velogénicos y mesogénicos. La prueba de índice de patogenicidad intracerebral del virus, en esta investigación, tuvo un índice de 1.14 que está en el rango de 1.0 a 1.5 y corresponde a virus mesogénicos considerados virulentos. Finalmente, el análisis filogenético del virus mostró que este virus se agrupa en el genotipo V de la clase II que corresponde a virus mexicanos.

Relevancia clínica. El virus de la enfermedad de Newcastle (VEN) está presente en los pollos y las gallinas de México desde los años 70’s, y los estudios filogenéticos sobre estos virus en los últimos años, muestran que la avicultura mexicana tiene dos linajes del genotipo v. Uno de ellos relacionado con la cepa Chimalhuacán que persiste en el país desde la década de los 70’s del siglo pasado y otro que ocasionó el brote en La Laguna en el año 2000 y que se le conoce como virus Torreón. El presente caso clínico refuerza la persistencia del virus de Newcastle en México y la vigilancia que se debe tener sobre la enfermedad en todo tipo de pollos y gallinas.

Newcastle disease in creole chickens: A pathologic and phylogenetic study

Case description. Ninety percent of a flock with 800 creole chickens, of 3 week of age, died within 72 h. Integral diagnoses of five chickens with depression, cachexy and serum secretion in the nasal and ocular mucous membranes, suggested that the chicken had a disease of high mortality such as the Newcastle disease or a highly pathogenic form of avian influenza.

Clinical findings. Bleeding from the trachea, proventriculus and cecal tonsils was observed. Histopathological analysis also revealed necrosis and bleeding in the bone marrow, thymus, spleen, bursa, cecal tonsils, trachea and bronchi. A severe hematopoietic hypoplasia with multifocal deposits of acidophylic material and cellular necrosis was observed in the bone marrow. Depletion of lymphocytes was observed in the thyme, spleen and tonsils, whereas the lymphoid follicles in the bursa were atrophied, and the bursa had an adenoidal shape with proliferation of the fibrous connective tissue and diffuse infiltration of lymphocytes. A large multifocal necrosis of lymphocytes was observed in some foils with lymphoid follicles. Together, the examination of the lesions at macroscopic and histological scales, suggested an acute and systemic necrosis, thus supporting a diagnosis of avian influenza or Newcastle disease.

Complementary laboratory tests: Tissues of the lung and trachea exhibited the most significant macroscopic lesions. The virus isolated from the trachea and analyzed with RT-PCR was positive for Newcastle disease. Sequencing of the gen F, suggested a 97 % similarity with the strain México/01/10 (JX974435) of Newcastle disease. Moreover, the virus had the same four basic aminoacids in the cleavage site of the portein F (RRQKR*F), as the velogenic and mesogenic pathotypes. The intra-cerebral pathogenic index found in this study was 1.4 within the range of 1.0 and 1.5, corresponding to the mesogenic viruses which are considered to be virulent. Finally, phylogenetic inference suggested that the virus was a close relative of the genotype V of the class II, corresponding to mexican viruses.

Clinical relevance. Newcastle disease virus (NDV) has been present in the poultry of México since 1970s, and phylogenetic studies suggest that two lineages of the genotype V circulate in mexican poultry farms. One of these strains is related to the strain Chimalhuacan that persist in the country since the 70s. The other one, Torreon virus, produced an outbreak of Newcastle in La Laguna in the year 2000. The present clinical case confirms the presence of NDV in México, and emphasizes the importance of epidemiological surveillance in all type of poultry farms.
